DEVELOPMENT OF TLC DENSITOMETRIC METHOD FOR THE QUANTIFICATION OF VALERENIC ACID IN VOLATILE OIL OF SELINUM VAGINATUM ROOTS

The present work was planned to develop and validate a simple, easy, and cost-effective method for the estimation of valerenic acid (VA) in volatile oil of Selinum vaginatum roots. Chromatographic estimation was performed using precoated silica gel 60 aluminium sheets (10 x 10 cm). Powdered roots of S. vaginatum (10 g) were hydrodistilled for 4 h in a Clevenger apparatus to obtain the volatile oil. The reference standard VA was resolved from the volatile oil of S. vaginatum roots using chloroform as mobile phase, scanned at 220 nm. The TLC densitometric method was found to be precise with good linearity (r2 > 0.996). The mean content of VA in the volatile oil was estimated to be 0.178% w/V. The developed method for the estimation of VA in S. vaginatum roots was validated using ICH guidelines and, thus, the observations conclude the method to be precise, accurate and reproducible.
